Abstract: Objective To identify the wood species of the detected plant materials involving in a traffic case.The rDNA-ITS sequence fragments of plant DNA barcode were analyzed for determining whether the bark-like materials collected from the left side of the tricycle were the same species as those carried on the agricultural vehicles. Method The plant DNA was extracted by the modified CTAB method. rDNA-ITS PCR amplification was performed and then the PCR products were cloned, sequenced, and the information of sequences was used for Blast analysis and phylogenetic tree construction. Result The adhesional substances on tricycle were originated from the Citrus sunki, while the plant shipped in the agricultural vehicles was Acer buergerianum (the variant species of JieShan Triangle), which was inconsistent with that of the bark on tricycle. Conclusion rDNA-ITS sequence fragments as plant DNA barcode could be applied for plant species identification.
摘要: 目的 以rDNA-ITS序列片段为植物DNA条形码对涉及一例交通命案的植物检材进行种属鉴定,判断三轮车左侧车体上提取的树皮样物质与农用车所运的树木是否为同一树种。方法 采用改良CTAB法提取植物DNA,先将DNA纯化,再进行rDNA-ITS的PCR扩增,然后对PCR产物切胶、回收,克隆、测序,测序结果进行BLAST分析,并构建系统发育树。结果 三轮车上粘附的树皮来源于柑橘(Citrus sunki),农用车所运的树木为界山三角槭(Acer buergerianum),两者非同一种属。结论 用rDNA-ITS序列片段作为植物DNA条形码对植物种属进行鉴定,可获得可靠的鉴定意见。
