Distribution of Five Sedative Hypnotics and Metformin in Hair by LC-MS/MS

Abstract: Objective To study the distribution of five hypnotics (diazepam, zolpidem, et al) and metformin in the hair samples of users, providing reference for characterizing the medication history. Methods The hair samples were segmented along the growth direction, ground and extracted. The content in each hair segment was determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS) and analyzed with the donors’ medication information. Results The distribution range of diazepam, alprazolam, clonazepam, and metformin in the hair samples corresponded to the duration of medication. The contents of analytes were related to multiple factors such as physical and chemical properties of the drugs, dosage, hair color, and individual metabolic differences. Conclusion The segmental hair analysis and drug distribution characteristics of real users can provide basic data support for the characterization of donors’ medication history in case investigation.

摘要: 目的 研究地西泮、唑吡坦等镇静催眠药物及二甲双胍在用药人员毛发中的含量分布特征,为毛发供体的摄药史刻画提供参考。方法 采集真实用药人员毛发,沿生长方向分段后研磨提取,采用液相色谱-串联质谱法(liquid chromatography-tandem mass spectrometry,LC-MS/MS)检测各段药物含量,并与供体用药信息进行关联分析。结果 地西泮、阿普唑仑、氯硝西泮、二甲双胍在用药人员毛发中的分布区间与用药时间存在良好对应关系,药物含量与药物理化特性、用药剂量、头发颜色及个体代谢等多重因素相关。结论 真实用药人员的头发分段检验和药物分布特征分析,能够为涉案毛发样本的供体摄药史刻画提供基础数据支撑。
