亲子鉴定中母亲嵌合体STR分型研究

STR Genotyping Analysis of Maternal Chimerism in a Parentage Testing Case

摘要: 目的 对1例三联体亲子鉴定中生母和女儿血样STR分型不符合遗传规律的原因进行分析。方法 利用GoldeneyeTM 20A试剂盒对被检父、女儿(小女儿)和生母血样进行基因分型检测及亲子鉴定,并进一步采集生母带毛囊的头发和口腔拭子及大女儿血样进行基因分型检测。结果 被检父与两个女儿均符合亲子关系,生母血样基因分型与小女儿基因分型排除亲子关系,与大女儿基因分型有2个基因座不符合遗传规律。生母头发中检出的基因分型与血样不同,且头发基因分型与两个女儿的基因分型均符合亲子关系。生母口腔拭子检出混合基因分型,包含其血样和头发的基因分型。 结论 本案例中母亲为罕见的嵌合体基因型,是两个女儿的生物学母亲。

Abstract: Objective To investigate the cause of the exclusion of a parent-child relationship between the biological mother and her daughters in a trio parentage test. Methods Genotyping and parentage testing were performed using the GoldeneyeTM 20A kit on blood samples from the alleged father, the daughter (younger daughter), and the biological mother. Additional samples, including the mother’s hair with follicles, buccal swab, and a blood sample from the elder daughter, were subsequently collected for further genotyping. Results The alleged father was confirmed to as the biological father of both daughters. However, the genotype obtained from the mother’s blood sample excluded her as the biological mother of the younger daughter and exhibited inconsistencies at two STR loci with the elder daughter’s genotype, violating Mendelian inheritance. A different genotype was detected in the mother’s hair sample, and the hair-derived genotype was consistent with her being the biological mother of both daughters. The mother’s buccal swab displayed a mixted genotype, comprising both the genetic profiles found in her blood and hair samples. Conclusion In this case, the mother was identified as a rare chimeric individual and is the biological mother of both daughters.
