Simultaneous Quantification of Canabinoids and the Major Metabolite THC-COOH in Urine Samples by LC/MS-MS

Abstract: Objective To develop a method for the simultaneous identification and quantification of THC, CBN, CBD and the major metabolite THC-COOH in urine samples. Methods Urine samples were hydrolyzed with sodium hydroxide, and deuterium-labeled THC-COOH (Δ9-d9-THC-COOH) was added as internal standard; then the solution was extracted with n-hexane-ethyl : acetate(9:1), dried, solved with 100μL acetonitrile, and analyzed by liquid chromatograph coupled to tandem mass-spectrometry (LC/MS-MS). Results The limits of detection for THC-COOH, CBN, THC, CBD were 0.2, 0.4, 1.0, 2.0ng/mL; THC-COOH was detected in the positive samples,with a concentration of 335.9ng/mL. Conclusion With this method, CBD,CBN,THC and their major metabolite THC-COOH in urine samples could be detected simply, quickly, sensitively and exclusively during toxic analysis.

摘要: 目的建立LC/MS-MS同时检测尿液中Δ9-四氢大麻酚(THC)、大麻酚(CBN)、大麻二酚(CBD)和大麻主要代谢物Δ9-四氢大麻酸(THC-COOH)的方法。方法尿液样本经碱水解,加入氘代四氢大麻酸(Δ9-d9-THC-COOH)内标,经V(正己烷):V(乙酸乙酯)=9:1提取,吹干,以100μL乙腈定容,利用LC/MS-MS方法进行分析。结果THC-COOH、CBN、THC和CBD的最低检测出质量浓度为0.2、0.4、1.0和2.0ng/mL;在阳性尿液中检出THC-COOH成分,质量浓度为335.9ng/mL。结论所建立的方法简便快速、灵敏度高、专属性强,可满足检测尿液中THC、CBN、CBD以及大麻主要代谢物THC-COOH的要求。
