Analysis of cytb and HVI in Mitochondrial DNA for Species identification

Abstract

Objective To study species identification of forensic biological specimens from the MtDNAsequence. Methods Fifteen species(human, cow, sheep, pig, fish, chicken, duck, dog,chimpanzee, monkey, panda, deer, lion, leopard and zebra) were concerned. Duplex amplification of cytb and HVI were carried out and the PCR products were analyzed by agarose gel electrophoresis. The cytb gene was sequenced and inquired. results Human and nonhuman samples could be simply distinguished by electrophoresis bands of cytb and HIVI PCR products, the species identification could be finished by inquiring cytb sequence matching on Internet(http://www.ncbi.nlm.nih.gov). Conclusion From the results obtained, it was shown that the mtDNAtesting system could be a valuable tool for species identification.

Objective To observe the shape and size of drip bloodstain on cement ground which dropped in different heights, speeds or height and speed. Methods Anticoagulation blood was dropped in static or movable conditions. The drip bloodstain was observed .The diameter was measured. The data were analyzed by special statistic software. Results In static conditions, the drip bloodstain was shaped round with stings on edge. There were linear relationship between the diameter and height, which could deduce linear formula. In walk, hurry walk, run and dash movable conditions, the drip bloodstain appeared as round or ellipse with stings on one side along direction of motion. Height or speed influenced the shape and size of drip bloodstain. Height and speed interacted on the shape and size of drip bloodstain. Conclusion The shape and size of drip bloodstain were useful to estimate the heights of bleeding, speeds and directions of motion.
