Screening for 132 Drugs in Blood by LC-MS/MS with Multiple-Reaction Monitoring

Objective To establish a screening system for simultaneous detection of drugs and toxic compounds in blood by LC-MS/MS. Methods Identification was based on the compound's ab-solute retention time, protonated molecular ion,and one representative fragment ion obtained by multiple reaction monitoring (MRM). The mass spectra library and criteria for identification were established. Results The MRM technique offers high sensitivity of SIS and structural information of scan,and low limits of detection (≤1ng/mL) for majority of the compounds (83%). Conclusion The method is sufficiently selective and sensitive in detecting the drugs down to therapeutic concentration levels and is therefore feasible in forensic and clinical toxicology.The method may also be adapted to other biological samples.
